Best Microscope for Pond Water and Microorganisms
Pond water is one of the most rewarding home-microscopy subjects. A compound microscope with transmitted light, fine focus and a mechanical stage can show algae, protozoa and other organisms in a simple wet mount.
The microscope type you need
Use a compound biological microscope because light must pass through the drop. A stereo microscope is useful for larger larvae or debris but does not provide the magnification needed for most single-celled organisms.
Magnification and contrast
Start at 40× or 100× to locate activity, then move to 400× for more detail. A 1000× oil-immersion objective is unnecessary for most pond observations. Lowering the condenser or partly closing the iris can improve contrast in transparent specimens.
Preparing a wet mount
Place one small drop on a clean slide and lower the coverslip at an angle to reduce bubbles. Begin at low power and use fine focus. Wash hands afterward and avoid collecting from polluted or hazardous water.
Mechanical stage and live movement
At 400×, tiny slide movements shift the field dramatically. A mechanical X–Y stage makes it much easier to follow a moving organism than pushing the slide by hand.
